The genes buttonhead (bt4 and PSp1 (The Dmsophila homologue of the human
Spl gene) encode zinc finger transcription facton known to play developmental
roles in the formation of the Dmsophile head segments and mechanosensory
larva1 organs. We find that these genes are expressed in the primordia of al1 ventral
imaginal discs (antennal, labial, leg and genital). Here we repon a novel function
of btd and D-Sp1 : they induce the formation of ventral cephalic and thoracic
imaginal disc primordia. These genes coexpress with Distel-less (010, heedcese
(hdc) and escargot (esg) in the lirnb primordium and theirfunction is necessary for
the activation of the latter genes. We also show that the Btd product is able to
adivate ectopically hdc and DI1 during embryogenesis. The expression of btd and
PSp1 in the embryonic thoracic segments requieres activity of wingless (wg). In
the abdominal segments the activating role of btdand DSp1 is suppressed by the
BX-C genes. Ectopic expression of btd during the development of the dorsal
imaginal discs (eyes, wings and halteres) induces formation of the corresponding
ventral stnidures (antennae and legs) and repress dorsal seledor genes as eyeless
(ey) and vestgial (vg). However, these ventral structures are not pattemed by the
morphogenetic signals present in the dorsal discs; the ceils expressing btdgenerate
their own signalling system, including the establishment of a sharp boundary of
engmiled (en) expression and the local activation ofthe wg and the decapentaplegic
(dpp) genes. Thus the Btd product has the capacity to induce the activity of the
entire genetic network necessary for ventral imaginal discs. This includes the
activation of antennal and leg patteming genes like homothorex (hth), dachshund
(dac) and 011.
