Síntesis y caracterización de nanopartículas magnéticas de ferrita de zinc para tratamientos combinados en biomedicina

Abstract
Las nanopartículas (NPs) magnéticas de óxido de hierro, como la magnetita y la maghemita, constituyen uno de los sistemas más prometedores en los campos de la biomedicina y la biotecnología. De entre los numerosos métodos de síntesis de estas NPs, el método electroquímico destaca por su reproducibilidad, robustez, versatilidad, bajo coste y por la posibilidad de obtener NPs de diámetro modulable en un amplio rango de tamaños, de entre 10 y 40 nm aproximadamente.
El estudio en esta Tesis Doctoral consistió, en primer lugar, en la optimización del método electroquímico para la obtención de una serie de NPs de ferrita de zinc de 12 nm de diámetro y diferente estequiometría, ZnxFe(3-x)O4 (0 < x <1). Para ello fue necesario el estudio del efecto de ciertos parámetros de reacción en las características del material sintetizado, tales como la intensidad de corriente aplicada a la celda electroquímica y la temperatura de reacción. El método de síntesis influye en gran medida tanto en las características estructurales como en las propiedades magnéticas de las NPs. Por este motivo, se realizó una caracterización exhaustiva de la serie de ferritas de zinc, definiendo las características del material obtenido mediante este método.
Las NPs obtenidas se recubrieron superficialmente con moléculas de dopamina, generando una plataforma para el posterior anclaje de moléculas biocompatibles, con el objetivo de aportar buena estabilidad coloidal y/o funcionalidad específica para su posterior empleo en biomedicina. En este trabajo las moléculas empleadas fueron los ácidos cítrico y fólico.
Posteriormente, se llevó a cabo un estudio toxicológico in vitro e in vivo (empleando el modelo animal Xenopus laevis) de las NPs. Los resultados indican que estas no son potencialmente tóxicas hasta concentraciones relativamente elevadas. Además, se obtuvieron datos esclarecedores sobre la absorción y el metabolismo de las NPs.
Por último, se evaluó la efectividad de las NPs sintetizadas para su posible empleo en tratamientos térmicos combinados contra el cáncer, concretamente en el tratamiento de tumores sólidos mediante hipertermia magnética y de terapia fototérmica. Los resultados obtenidos para las NPs estudiadas muestran un bajo calentamiento cuando se aplica un campo magnético alterno (SAR), en tratamientos de hipertermia magnética. Por el contrario, las NPs mostraron altos valores de eficiencia de conversión fototérmica, siendo posible su modulación mediante la variación de la composición química de las NPs. Magnetic iron oxide nanoparticles (NPs), such as magnetite and maghemite, constitute one of the most promising systems in the biomedicine and biotechnology fields. Among the numerous methods of synthesis of magnetic ferrite NPs, the electrochemical method stands out for its reproducibility, robustness, versatility, low cost, green chemistry and the possibility of obtaining NPs of tuneable diameter in a wide range of sizes, between 10 and 40 nm, approximately. The study carried out in this Doctoral Thesis consisted, firstly, in the optimization of the electrochemical method to obtain a series of zinc ferrite NPs of 12 nm in diameter and different stoichiometry, ZnxFe(3-x)O4 (0 <x <1). For this purpose, it was necessary to study the effect of certain reaction parameters on the characteristics of the synthesized material, such as the intensity of the current applied to the electrochemical cell and the reaction temperature. The synthesis method greatly influences the structural characteristics such as the magnetic properties of the NPs. For this reason, an exhaustive characterization of the series of zinc ferrites was carried out, defining the characteristics of the material obtained by the electrochemical method.
The obtained NPs were coated with dopamine molecules, generating a platform for the subsequent anchoring of biocompatible molecules, with the aim of providing good colloidal stability and/or specific functionality for their subsequent use in biomedicine. In this work, the molecules used were citric and folic acids.
Next, a toxicological study was carried out in vitro and in vivo (using the animal model Xenopus laevis). The results indicate that NPs are not potentially toxic up to relatively high concentrations. In addition, clarifying data were obtained on the absorption and metabolism of the NPs.
Finally, the effectiveness of the synthesized NPs for their possible use in combined thermal treatments against cancer was evaluated, specifically in the treatment of solid tumors using magnetic hyperthermia and photothermal therapy. The results indicated that the studied NPs are not suitable for magnetic hyperthermia treatments, due to their low SAR values. On the other hand, the NPs showed high values of photothermal conversion efficiency, being possible their modulation by means of the variation of the chemical composition of the NPs.
