Abstract
El área tegmental ventral (VTA) se caracteriza por su heterogénea parcelación citoarquitectónica, la diversidad de sus proyecciones y la riqueza bioquímica de sus neuronas, las cuales son mayormente dopaminérgicas, entremezcladas con otras menos numerosas gabaérgicas o glutamatérgicas, e incluso con neuronas que contienen combinaciones de estos neurotransmisores. Los principales sistemas de proyección del VTA son el mesocortical y mesolímbico, principalmente dopaminérgicos, cuya disfunción está implicada en la fisiopatología de la esquizofrenia y de trastornos adictivos. En esta Tesis Doctoral se han estudiado las proyecciones eferentes de subdivisiones concretas del VTA del ratón para determinar las posibles diferencias funcionales de estos territorios mesencefálicos. Para ello, se realizó un estudio poblacional de las proyecciones eferentes de varias subdivisiones del VTA usando microdepósitos de amino dextrano biotinilado y, después se llevó a cabo la transfección de 30 neuronas únicas ubicadas en distintas subdivisiones del VTA con el vector Sindbis-pal-GFP y la reconstrucción completa de sus axones, para examinar sus patrones de ramificación. En estas neuronas se analizó con microscopía confocal su fenotipo dopaminérgico usando inmunofluorescencia para la tirosina hidroxilasa y la longitud de axón de las terminaciones axonales se estimó con estereología. Según su patrón de arborización axonal se diferencian dos tipos de neuronas de VTA: las que proyectan al prosencéfalo y las que proyectan al prosencéfalo y al tronco del encéfalo. El primer tipo se divide en cuatro subtipos según sus principales dianas de inervación: neuronas mesocorticolímbicas (proyectan a la corteza cerebral y al prosencéfalo basal), neuronas mesocorticales (inervan la corteza cerebral), neuronas mesolímbicas (inervan estructuras límbicas prosencefálicas) y neuronas mesoestriadas (inervan el caudado-putamen). No se ha observado una topografía selectiva de ninguno de estos tipos neuronales en las subdivisiones de VTA estudiadas. La diversidad de conexiones del VTA y la riqueza de las arborizaciones axonales de sus neuronas demostradas en esta Tesis Doctoral es consistente con el hecho de que neuronas del VTA modulan la actividad de grupos específicos de estructuras prosencefálicas y troncoencefálicas. The Ventral Tegmental Area (VTA) is characterized by its heterogeneous cytoarchitectonic subdivisions, the diversity of its projections and the biochemical variety of its neurons, which are mostly dopaminergic, mixed with other, less numerous, gabaergic or glutamatergic ones, and even with neurons that contain several of these neurotransmitters. The main projection systems of the VTA are the mesocortical and the mesolimbic ones, largely dopaminergic, the dysfunction of which is involved in the pathophysiology of schizophrenia and addictive behaviors. In this Doctoral Thesis, we have studied in the mouse the efferent projections of specific subdivisions of the VTA, to determine the possible functional diversity of these VTA mesencephalic territories. For this purpose, the axons of small populations of VTA cells were labeled with iontophoretic microdeposits of biotinylated dextran amine, and then the axons of 30 VTA single neurons located in different VTA subdivisions were labeled and analyzed by means of single-cell transfection with the Sindbis-pal-GFP vector to define their patterns of branching. The dopaminergic phenotype of these single-labeled neurons was tested by double-labeling for tyrosine hydroxylase immunofluorescence and the length of terminal axonal arbors was estimated by stereology. Depending on their axonal branching pattern, two main, markedly different VTA cell morphologies were observed: 1. neurons with a single main axon targeting only forebrain structures, and 2. neurons with multibranched axons targeting both the forebrain and the brainstem. Moreover, four “subtypes” could be distinguished among the first type of neurons based on their projection targets: mesocorticolimbic neurons (projecting to neocortex and basal forebrain), mesocortical neurons (innervating the neocortex), mesolimbic neurons (innervating forebrain limbic structures), and mesostriatal neurons (targeting caudate-putamen). None of the neuronal types described in this study on the basis of their axonal branching patterns is distributed in a manner that correlates with the VTA subdivisions analyzed here. The observed diversity in the connections and in the wiring architectures of the VTA neurons, demonstrated in this Doctoral Thesis, is consistent with the notion that different VTA cell subpopulations modulate the activity of specific sets of prosencephalic and brainstem structures.
